Designed for isolating high-quality genomic DNA from soil samples (up to 250 mg soil per extraction).
Kit Components and Descriptions:
The kit comes in a compact box with labeled vials/bottles. Each kit makes ~50 extractions (scalable). Components are divided into consumables (used per extraction) and reusable items.

- L Buffer1 (12 mL vial)
- beta-mercaptoethanol
- LB2 (12 ml vial)

Usage Protocol:
This is a step-by-step guide for one extraction. Time: ~1-2 hours. Requires 250 mg soil sample.
1. Sample Preparation: Weigh 250 mg soil into a 2 mL tube. Add 500 µL L Buffer1 + 75 µL L Buffer2+ 5 µL Proteinase K. Vortex vigorously for 10 min.
2. Lysis and Incubation: Incubate at 65°C for 30 min (with occasional vertexing). Cool to room temp.
3. OB: Add 500 µL OB. Vortex 30 sec, centrifuge at 10,000 rpm for 5 min. Transfer upper aqueous phase (~400 µL) to a new tube.
4. BB: Add 500 µL BB to the aqueous phase. Mix and load into spin column in a collection tube. Centrifuge 1 min at 10,000 rpm. Discard flow-through.
5. Washing: Add 500 µL W buffer 1 to column. Centrifuge 2 min, discard flow-through. Repeat with 500 µL W buffer 2. Centrifuge again to dry (2 min).
6. Elution: Place column in a new collection tube. Add 50-100 µL DI water or elution buffer. Incubate 1 min, centrifuge 1 min. Collect eluted DNA.
7. Storage: Store DNA at -20°C. Quantify with NanoDrop or PCR.
