TOS Propionate Agar (TPA) is a medium for the direct detection of viable Bifidobacteria. The method is applicable to milk products such as fermented and non-fermented milks, milk powders, infant formulae, and starter cultures where these microorganisms are present and viable, and in combination with other lactic acid bacteria.

In combination with MUP, it allows the direct detection of viable Bifidobacteria.

Application

-M-17 Broth is a selective medium.
-Specifically designed for the cultivation of Streptococcus thermophilus.
-Widely used in yogurt testing.
-Applied for the enrichment of S. Thermophilus in the dairy industry.

Preparation

Suspend 54.7 g of dehydrated medium in 950 ml of distilled water (use 1 liter for non-selective medium). Heat to boiling to dissolve the medium completely. Sterilize by autoclaving at 115 °C for 15 minutes.
For selective isolation of Bifidobacteria, add the contents of two 25 ml vials of Lithium Mupirocin Supplement. Mix carefully to avoid air bubble formation and dispense as desired.

Note: This medium is sensitive to heat; excessive heat treatment may adversely affect its properties.
